Ингибиторы ферментов репарации ДНК как прототипы лекарственных препаратов

Аннотация:
Стабильность генома высших эукариот во многом обеспечивается функционированием систем репарации ДНК. Активность этих систем может обусловливать устойчивость раковых клеток к химио- и радиотерапии, в связи с чем ферменты - участники и регуляторы систем репарации выступают как многообещающие терапевтические мишени. Рассмотрены современный уровень и перспективы исследований ингибирования трех ферментов репарации ДНК, а именно поли(АДФ-рибозо)полимераза 1 (PARP1), поли(АДФ-рибозо)полимераза 2 (PARP2) и тирозил-ДНК-фосфодиэстераза 1 (Tdp1), как фармакологических мишеней.
