Abstract:

Pigs are a recognized test system for conducting biomedical research. Data from the latest meta-analysis showed that the required level of analgesia and anesthesia is rarely provided to pigs. Propofol and isoflurane are attractive for their rapid withdrawal from anesthesia on the one hand and the possibility of prolonged administration and retention of anesthesia on the other. The addition of tramadol to these drugs improves analgesia in polymodal anesthesia.

The purpose of this study is to study the effect of the tramadol-propofol (bolus)-propofol (infusion) regimen, in comparison with the tramadol-propofol (bolus)-isoflurane combination on the basic physiological parameters in dwarf pigs.

The study was approved by the local Bioethics Commission No. 2.50/24. 10 male and 10 female dwarf pigs aged 1 to 3 years were used. In both cases, tramadol was administered intramuscularly (i.m.) at a dose of 10 mg / kg, followed by intravenous propofol at a dose of 6 mg / kg after 35-40 minutes. Further, in the first case, propofol was administered at a constant rate of 8 mg / kg/hour, and in the second case, a mask supply of isoflurane was used with a flow rate of 2 l/min and a concentration of 1.5% in an air mixture with 40-93% oxygen. The physiological parameters were measured after the 80th minute of propofol (IPS) and isoflurane administration.

It was found that both anesthesia regimens can provide controlled anesthesia as early as 15-40 seconds after bolus administration of propofol. Anesthesia continues throughout the duration of propofol (infusion) or isoflurane (inhalation) administration. After stopping the supply of anesthetics, reversion after propofol occurs after 26-29 minutes, whereas after stopping the supply of isoflurane, it occurs already at 12-15 minutes. The use of both anesthesia regimens reduces the saturation level, which requires monitoring and additional oxygen supply. When using them, there is a statistically significant reduction in the frequency of respiratory movements, while the use of a combination with isoflurane causes the most severe decrease compared with a combination with propofol. In addition, both anesthesia protocols reduce blood pressure and heart rate, have a similar effect on electrocardiogram readings, and also have no effect on hematological blood analysis. The combination with isoflurane has no effect on the biochemical picture of the blood, whereas the scheme with propofol significantly reduces the level of alkaline phosphatase.

The results obtained can be useful in preclinical studies for performing manipulations of mild and moderate severity.
