Abstract:

Currently, when developing intranasal drug delivery systems for local/systemic effect, it is necessary to evaluate their cytotoxicity and transport of substances through the nasal epithelium, for which the RPMI 2650 cell line is actively used in in vitro experiments.

The aim of the work was to evaluate the formation of a cellular monolayer by the content of proteins of intercellular contacts ZO-1, occludin, claudin-1, E-cadherin of the RPMI 2650 cell line in the dynamics of cultivation from day 1 to day 8.

The study of RPMI 2650 elements using light microscopy showed that by the 5th day of incubation, the cells form a monolayer, which thickens by the 8th day of observation. At the same time, the cells formed dense intercellular contacts, as evidenced by the dynamics of an increase in the relative number of proteins ZO-1, occludin, claudin-1, and E-cadherin.

RPMI 2650 cells cultured in a MEM nutrient medium with the addition of 10% FBS, 2 mM L-glutamine, 100 U/ml penicillin and 100 mg/ml streptomycin form a monolayer on the 5th day of cultivation, which is compacted by the 8th day of observation. The formation of a monolayer is accompanied by an increase in the relative number of proteins of intercellular contacts ZO-1, E-cadherin, claudin-1, occludin on days 5 and 8 of cultivation, which allows using this cell line to study the transport of exogenous substances in vitro.
