Method for detecting perineural and intraneural invasion in breast cancer as a prognostic factor of tumor progression

Abstract:

Despite the fact that perineural invasion (PNI) is a generally recognized metastatic process in malignant tumors of various locations, in cases of breast cancer to date, PNI remains an insufficiently studied way of metastatic spread.

Aim of the work – to develop an easy-to-use and reliable pathological diagnostic method for visualizing perineural and intraneural tumor invasions in invasive ductal breast cancer of a nonspecific type.

The study was performed on archival material from 36 patients with invasive breast carcinoma of nonspecific type G2–G3 (IC NST). The study is made in two stages: the first stage is histological diagnosis by staining preparations with hematoxylin and eosin, with the obligatory assessment of clusters of tumor cells, with spread into the perineural spaces, as well as directly into the nerve trunk; The second stage is the IHC technique with the simultaneous use of neuronal (for example, S100) and PanCK (for example, AE1/AE3) markers, with different color mark (for example, S100 – pink mark, AE1/AE3 – brown mark). Thus, an obvious contrast between tumor tissue and neural tissue is achieved, which provides high accuracy in assessing perineural and intraneural tumor invasion.

An original pathological diagnostic algorithm for analyzing the metastatic potential of invasive ductal breast cancer of a nonspecific type is proposed, characterized by a new method of visualizing perineural and intraneural tumor invasion.
