Abstract:

Recently, in medicine, special attention has been paid to methods based on non-invasive approaches, including the use of so-called "therapeutic" enzymes that provide a pronounced effect at low concentrations and physiological conditions. Such enzymes include collagenases - endopeptidases that catalyze chemical processes and destroy peptide bonds in collagen. Collagen is the main structural protein of the intercellular matrix which plays an important role in physiological and pathological processes in the body. In this regard, protein hydrolyzing enzymes have significant potential for use in medicine. In addition, collagenases can be used in the food industry, pharmacy, to solve environmental problems. All of the above determines the relevance of the search for new producers of collagenolytic proteases and optimization of enzyme synthesis conditions. The aim of the work was to study the effects of cultivation conditions on the collagenolytic proteases secretion by Aspergillus fumigatus. The object of the study was mycelial fungus Aspergillus fumigatus F 22 from the VILAR microorganisms collection. Submerged cultivation was carried out using a liquid modified medium Chapek Dox with partial replacement of sucrose for collagen in flasks or fermenter. The concentrations of protein and collagenolytic activity were determined in the culture medium filtrates. The maximum secretion of collagenolytic proteases was observed at a concentration of 0.5% sucrose and 1.5% collagen in the nutrient medium. Among the studied concentrations of spores, we detected, that the maximum collagenolytical proteases production in submerged cultivation were with 5x106 spores/ml. Effect of carbohydrate sources and nitrogen sources on the collagenolytic enzymes production by A. fumigatus has been studied as well. It is shown that the growth rate and the biomass weight of A. fumigatus depend on the inoculation methods. It was found that use of 5% mycelium provides maximum collagenolytic proteases secretion by A. fumigatus. Based on the results obtained, the method of inoculation with 5% mycelium was chosen for cultivation in the fermenter. The growth rate and production of collagenolytic proteases were compared during cultivation in flasks and in the fermenter. It is shown that there are no statistically significant differences in these indicators for the two cultivation methods. The results obtained are important for a new A. fumigatus based collagenolytic proteases production technology development.
