Abstract:
Now extensive researches are conducted in usage of carbon nanotubes in diagnosis and medicine. It is few known about the influence of nanotubes on nervous and immune system of organism.
The aim of the study was to investigate the cytokines synthesis (mRNA levels of interleukins (IL)-1beta, -2, -4, -5, -6, -8, -10, -13, -17, -18, tumor necrosis factor (TNF)-alpha, interferon (IFN)-alpha, IFN-beta, IFN-gamma by the method of RT-PCR and expression of c-fos by the method of immunohistochemistry in the rat brain under intranasal administration of single-walled carbon nanotubes (SWCN). In the experiment we used a paste of SWCN, provided by Krestinin A.V. (Institute of problems of chemical physics RAS, Russia). We used two groups of rats: experimental (n=6, intranasal administration of SWNT in dilution 1:1000) and control (n=6, intranasal administration of saline at the same volume). A day after single intranasal administration of SWNT the rat brain was taken for study: a right half divided on anterior and posterior parts was used for immunological study, and left half - for immunohistochemical study. According to immunological study, SWNT activated gene expression of IFN-gamma and TNF-alpha, but inhibited transcription of IL-13 and IL-17 in the posterior part of brain. At the same time in anterior part we detected the activation of mRNA IL-10 and suppression of the synthesis at the transcriptional level of IFN -β, IL-1β, -5, -6, -8 and TNF-a. Although in the blood of the same rats under the influence of SWCN gene expression of IFN-γ, IL-8, -18 and TNF-α was activated that indicates the activity of Th1 and macrophages. At the same time the transcription of IFN -β, IL-1β, -6 was inhibited. Probably Th1-type immune response activated in the posterior part and Th2-type activated in anterior part of brain. According to immunohistochemical study, administration of SWNT did not changed c-fos expression compared to control, except olfactory bulb, where we observed the increase of c-fos expression (p=0.12 - tendency). Thus, intranasal administration of SWNT did not influenced on brain activity. We assume that SWNT can be used for creating the new effective methods of brain diseases treatment.
