Abstract:

Continuous ambulatory peritoneal dialysis (CAPD) is an effective and widely used method of renal replacement therapy. Effluent dialysate drained from the peritoneal cavity contains multiple substances, e.g., advanced glycation end products (AGE) and indoxyl sulfate, which are clinically significant biomarkers. The aim of this research is to investigate the possibility of direct determination of these biomarkers in peritoneal dialysate and urine using rapid chromatographic separation, spectrophotometry and UV-Visible fluorescence spectroscopy. It was demonstrated that the absorption and fluorescence spectra of chromatographic fractions of dialysate and urine show characteristic features of proteins, AGE, urea, creatinine and indoxyl sulfate, which reveals the potential possibility of their direct quantitative determination by optical spectroscopy. The compact sensor based on fast proteins and metabolites liquid chromatography, supplemented with a fluorescence detector, used in this research for the separation of biomolecules, can potentially be used for rapid diagnostics of inflammatory processes and assessment of protein losses in patients on CAPD in outpatient settings.
