Abstract:
At present research we have studied the influence of gap junction blocker carbenoxolone on the distribution of neuronal gap junction contained connexin 36 (Cx36) after cerebral ischemia.
At present work we have produced ischemia by photochemical thrombosis in somatosensory cortex of rat brain. All rats were divided into 2 groups: active control (with cerebral ischemia) and experimental group (with cerebral ischemia and carbenoxolone treatment, dosage: 1mg/kg). Next day their brains were taken for morphological research. Immunohistochemical staining were produced by standard method using primary polyclonal rabbit antibody to Cx36. In the core, penumbra and intact area of the neocortex we have calculated the average number of gap junctions per one cell. We have obtained that in rats under carbenoxolone treatment after cerebral ischemia the number of neuronal gap junctions was increased in the core and penumbra compared with control rats. We assume that the increase of neuronal gap junctions may be due to adaptive reaction in response to carbenoxolone injection.
