Abstract:
The plant flavonoids are widely used as the antioxidant agents. We also know their antiallergic, anti-inflammatory, lipid-lowering and some other biological functions [1]. We investigated five synthesized thiazole derivatives of isoflavone obtained at the Department of Organic Chemistry of Kiev University. These new thiazole analogues of isoflavone with substituents at position 1 and 3 of flavonoids FL-7, 9, 12, 12a, 12b, have very low toxicity [2]. Experiments were conducted on three types of animals kept on atherogenic rich cholesterol - hypercholesterolemic diet. Control animals received standard diet.
Hypercholesterolemic diet in animals leads to an increase in cholesterol and triglycerides content in the blood serum and liver, and reduces antiatherogenic lipoprotein fraction - high-density lipoproteins (HDL). Supplementation of isoflavones to such animals in a dose 300 mg per kg body weight increases the concentration of decreased antiatherogenic HDL [3]. The separation of lipoproteins by ultracentrifugation or by electrophoresis in polyacrylamide gel revealed the recovering of HDL cholesterol, which was not determined in hypercholesterolemic diet without using of flavonoids. The highest activity of studied isoflavones manifested FL-12, the lowest FL-7. In experiments in animals with experimental hyperlipidemia - rats, guinea pigs and rabbits, we established a higher lipid-lowering activity of thiazole derivatives of isoflavone on impaired lipid and lipoproteins metabolism. The most lipid-lowering action possessed preparation FL-12 (picture). The thiazole derivatives of isoflavone seem to be promising as a new hypolipidemic and antioxidant drugs.
