Dose-dependent effect of dopamine on inotrope infarction 21-day rats

Abstract:
It is known that the autonomic nerve endings found not only the base but also additional mediators. Thus, sympathetic nerve endings contain many dopamine  the precursor synthesis of catecholamines, which acts on target cells via specific as dopamine receptors , and through the cross-binding to α- and β-adrenoceptors [3]. The features of the topography of the receptor structures to mediators of peptides in the sinoatrial node of rats, where the core functionality of the sinoatrial node of the heart in the central part of the largely impoverished content primary neurotransmitter receptors of the autonomic nervous system [2]. Involvement in regulating dopamine activity of the heart has been insufficiently studied, particularly in a dose dependent legandy, there are only few studies. The aim of this work was to study in vitro dose-response effect of dopamine on the background of β-adrenoceptor blockade with propranolol, α-adrenoceptor antagonists phentolamine and d-receptors droperidol (production Sigma) on inotropic cardiac function 21-day-old rats. Learning dopamine concentration range 109-105 M. Isometric contraction strips atrial and ventricular myocardium were recorded on the installation «Power Lab» (ADInstrumets Australia). In 21-day-old rats dopamine in a concentration of 109 M has a positive inotropic effect on the myocardium of the atria and ventricles. Dopamine in a concentration of 10 8M increases the force of contraction in the ventricles, in the atria reduces contrary. Further determination of dose response showed that dopamine concentration of 107 M, 106 M, 105 M and causes only the negative inotropic effects on the myocardium as atrial and ventricular [1]. On the background of the blockade d-receptors in 21-day-old rats dopamine in a concentration of 109 M, 108 M, 107 M causes negative inotropic response. Backlash in the ventricles and is stored at a concentration of 106 M. Dopamine concentration of 105 M strips increases the force of contraction of the atria and ventricles of the myocardium. Dopamine exploring all concentrations (109 - 105 M) on the background of β- adrenoceptor blockade with propranolol reduces the force of contraction of the atrial myocardium and ventricular myocardium. When α-adrenoceptor blockade of phentolamine in 21-day-old rats at a concentration of dopamine, 109 M, 108 M, 107 is a unidirectional response, i.e. decrease in the force of contraction of the atria and ventricles of the myocardium. The highest negative response was in the atria. Dopamine concentration 106 M strips increases the force of contraction of the atrial myocardium, and the force of contraction of ventricular myocardium reduces strips. A lower concentration of dopamine 105 M causes an increase in the force of contraction of the atria and ventricles of the myocardium. Consequently, the positive inotropic effect of dopamine in a concentration of 10-9M implements via dopamine, a concentration of 105 M  via β-adrenergic receptors.
