Abstract:
The influence of acoustic (ultrasonic) waves on leukocytes and on the output of practically important metabolites was studied. It was explored the possibility of changing the mode of receiving interferon (IFN). The drug quality has been controlled by biochemical methods. IFN was isolated and purified biologically. 3 minute ultrasonic exposure stimulates the release of IFN. Its activity increases. The most optimal US intensity dose was 0.01-0.2 W/cm2 with an exposure of 2-4 minutes. Then stimulation decreased and the stimulating effect was kept constant at the US dosage range of 0.4-0.7 W/cm2. Increase of the US time treatment for more than 4 minutes, and the US intensity above 0.7 W/cm2 led to a sharp decrease of IFN activity. It could be done the conclusion that the optimal level of US intensity for "waste" cells is several times lower than the intensity of the US used to stimulate the production of IFN by the induced cells and was 0.05-0.2 W/cm2, the exposure time varied from 30 seconds to 2-4 minutes. As a result, we managed to get up 22-30% and more of the IFN additionally and to use the donor blood more efficiently. The use of analysis of variance was tested for predicting the yield of protein production. The biological assessment of the quality produced IFN in animals by the ratio of the number and mobility of the protein fractions of blood serum was conducted. IFN activity and lack of its toxicity was controlled using luminescent bacteria. The biological effects of the formulation produced has been tested in animals Biochemical and clinical usefulness of these formulations in addition shows a lack of devastating denaturing of action of the intensities. Therefore, further study of the possibility of non-invasive management of the functional state of the cells with the help of ultrasonic waves is promising, relevant and theoretically meaningful.
