Abstract:
Collagenase are promising drugs for therapy. Getting them from animal tissues constrained by the small content, but it is possible to obtain enzymes, using microorganisms. We have shown previously that micromycetes A. flavus secretes two collagenases with different properties and examined the factors that influence the quantitative and qualitative composition of the secreted enzymes. However, all works were carried out using low volume (in flasks with 100 ml of medium), that is not enough for pre-clinical research and drug development. The aim of the research was to study the possibility of obtaining collagenases A. flavus with the use of fermentater.
Deep cultivation of A. flavus spent in two fermentaters (№ 1 and № 2) MD-250-2,6, «Marubisi» volume of 2,7 l with 1 l modified nutrient environment Czapek. Fermentation apparatuses differed coefficients of mass transfer: 21 h-1 (№ 1) and 30 h-1 (№ 2). Seeds served 25 ml 7-day suspension dispute A. flavus (3,45x107 spores/ml) grown on agar medium Czapek.
Earlier accumulation of the protein occurs in a apparatus № 2, which correlates with the speed of utilization of sucrose. In apparatus № 2 full use of carbohydrate happens to 48 hours, while in № 1 - the residual amount of sucrose is present up to 66 hours. Proteolytic activity of cultural liquid grew as the use of sucrose in the process of fermentation, reaching a maximum of 48 (apparatus № 1) and 66 hours (apparatus № 2), and then declined. The protease, excreted by A. flavus, had collagenolytic activity comparable to those under cultivation in flasks on a shaiker. However, the time required for the secretion of enzymes substantially reduced when using fermentater.
Thus, as a result of the conducted research demonstrated the possibility of scaling the process of cultivation A. flavus - producent collagenases.
