Abstract:
Now large number of vegetative raw materials containing Ascorbic acid is applied both in modern medical practice and as biologically active food additives. Ascorbic acid is contained in many vegetative objects. Quantitative determination of ascorbic acid in rosehips is carried out by the way of titration using 2,6-dichlorophenolindophenolate sodium. In most cases evaluation of quality of vitamin-containing raw materials is carried out using other groups of biologically active substances.
The technique of quantitative fluorimetric determination of ascorbic acid in rosehips is offered. The technique includes its preliminary oxidation by copper acetate solution with subsequent condensation of dehydroascorbic acid with o-phenylenediamine. Sample preparation and testing conditions were developed. Validation assessment of the method and comparison with titrimetry method were carried out. The analysis of the obtained test results in view of the following factors: linear dependence of fluorescence intensity on concentration of the substance being determined in the solution, analytical span of concentrations, specificity and accuracy (correctness and precision) of the method - has shown that other biologically active and ballast substances do not interfere with determination of ascorbic acid. Validation assessment demonstrates reproducibility and specificity of the technique offered. Approbation of fluorimetric technique using rosehips makes it possible to recommend the developed technique for determination of ascorbic acid in vegetative raw materials and for inclusion in the standard documentation for vegetative raw materials.
