Abstract:
Qualitative and quantitative composition of carbohydrates in fruits of Phellodendron amurense Rupt. is not well known. However knowledge of qualitative and quantitative composition of sugars is important because carbohydrates help active substances of phytopreparations to penetrate in to the cells of human body and enhance their action.
Objective of the study was to develop a method for qualitative determination and assay of carbohydrates and establishing their qualitative composition and quantitative content in fruits of Phellodendron amurense Rupt.
Object of the study was fruit picked up in September dried in aerated protected from light place at humidity 14 %.
In the prepared raw material qualitative and quantitative content of carbohydrates was determined by steps. On the first step sugars were identified by qualitative reactions, on the second step sugars were identified by TLC, on the third step qualitative and quantitative composition of carbohydrate was determined by HPLC method.
Free sugars content was assayed by HPLC technique on Liena NH2 column 4.6×250 mm, mobile phase - acetonitrile: water (70:30). Content of bound sugars was assayed by Capillary Electrophoresis method using Applied Biosystem 273 T.
From free sugars both fructose and glucose were present. Fructose content was from 15,7 % to 16,1 %, glucose - from 15,1 % to 18,3 %. Bound sugars were represented by glucose and xylose. Glucose content was from 1,1 % to 1,8 %, xylose - from 0,83 % to 0,95 %.
