Results of development of method of identification and genotyping of microorganisms causing the infections of respiratory tract

Abstract:
Despite of the gained results, the diagnostics of respiratory tract infections is still remaining the significant problem of the modern public health. It is connected with the lack of the appropriate methods of identification of these pathogens. Objec-tively, the diversity of these fastidious species is another and even more important reason. The aim of our research was to develop new method of identification and genotyping of respiratory tract pathogens in case of Streptococcus pneumonia as the most frequent respiratory bacterial pathogen. Methods. We studied non-coding region of Streptococcus pneumoniae DNA between SP_1922 and SP_1923 which was marked as NCR (non-coding region). There were defined such peculiarities of this region as the availability of species spe-cific ply gene, the appropriate size of amplicon and the statistically approved variability of restricted fragments. Results: Non-coding region (NCR) of DNA of S.pneumoniae is species specific area of genome and have the peculiarities al-lowing to use as the optimal diagnostic target for identification of Streptococcus pneumoniae. The main features of the NCR fragment is the appropriate size of amplicon (1182 bp), amplification of variable are of genome, the stability of flank regions (SP_1922 hypothetical protein) and SP1923 (pneumolysin). Conclusion: non-coding genome is still remains the perspective source of the diagnostics targets for respiratory tract pa-thogens. Terminal restriction fragments length polymorphism also is the appropriate way to develop the new method of intraspecies differentiation.
