Abstract:
The actuality of microbiological monitoring for the detection of pathogens in the environment objects demands the development of high sensitive and specific piezoelectric biosensors (PB) technology.
The test-systems construction on the base of the PB for detection of plague, brucellosis, tularemia pathogens with the help of gravimetric flow injection analysis and conduction of experimental laboratory trials was the main aim of this investigation.
For realization of the main aim of this work the specific immunoglobulins against used pathogens were applied for the PB construction. The quartz crystal resonators ДМ-7 with the gold electrode as well, as quartz analyzer «CPNA-330» and flow cell implement produced by «NPF CJSC ETNA» (Russia) and peristaltic pump «LKB 2132» (Sweden) were used in this experiments. Also, ourselves created adjustments and implements were used for the BP preparation.
The experimental series of test systems for the detection of pathogens plague, brucellosis and tularemia on the base of PB have been prepared. Each test system includes three PB, pathogen cells suspension 1,0×109m.c./mL, 0,01 M PBS (dried salts), ampules file, manual. Еxperimental laboratory tests were conducted to approve the suitability of the above shown test systems.
Advantages of the application PB test-systems in the gravimetric flow injection analysis were the high sensitivity up to 1,0×103 1,0×104 m.c./mL, the absence of cross-reactions with the heterologous suspension of strains, the speed setting about 15 minutes and the informative results of analyzes as tables and graphics data in computer. According the experimental data test system have shown conformity to the claim of diagnostic preparation instruction.
According to the material of this article the invention claim № 2012114858 was considered by Rospatent and the positive decision was taken by FIPS on 20.11.2013 г.
