Effect of dopamine on myocardial inotropy immature rats

Abstract:
Investigated the effects of dopamine at concentrations 10-9 -10-5M on the myocardial contractile activity of the rat 21-day age. In analyzing of the isometric contraction strips infarction it has been found that the dopamine in a concentration of 10-9 M increases the force of myocardial contraction. Further determination of dose response showed that dopamine in a concentration of 10-7 М, 10-6 М, 10-5 М, has only a negative inotropic effects on the myocardium of the atria and ventricles. Adding dopamine during blockade of α-adrenergic receptors by the phentolamine on 21-day-old rats resulted in an increase of the force of contraction of the myocardium at a concentration of 10-5 М. All subsequent concentration caused decrease of the strength of myocardial contractility. Dopamine on the background of β-adrenoceptor blockade with propranolol retains negative inotropic effect. On the background of the blockade d-receptors of 21-day-old rats the dopamine positive inotropic reaction causes at concentration of 10-5 М. Consequently, the positive inotropic effect of dopamine in a concentration of 10-9 М realizes through the dopamine, in a concentration of 10-5 М - through the β-adrenergic receptors.
