Effect of afobazol on the postnatal development of the offspring of rats exposed to prenatally exposure combustion of peat smoke in vivo

Abstract:
Objective To evaluate the postnatal development and production of conditioned reflexes of offspring obtained from rats exposed to peat smoke in the prenatal period and correcting influence of afobazol on manifestations of observed deviations. Materials and Methods On white rats daily from 1 to 20 days of pregnancy, exposed peat smoke within 24 minutes from 4 consistently burning paper tubes filled with 70% peat , 30 % wood pulp. Before inhalation treatment rats were administered per os afobazol at doses of 1 mg/kg and 10 mg/kg, postnatal female-s afobazol received a dose of 200 mg/kg from the 1st to the 20th days of feeding offspring. The study of male rats (60 days old) exposed peat smoke prenatally showed displacement of locomotors activity in the direction of increasing horizontal displacement and violation of extinction of the orienting-exploratory behavior with repeated (5 days) presentation of the test «open field» (OF). This indicates the presence of cognitive impairment (associative learning) and increased emotional reactivity in this group of animals. Influenced afobazol in the offspring of rats exposed to peat smoke in the test OF, there was a decrease of horizontal (center and 2/3 of the field) and vertical (stand) locomotors activities (p ≤ 0,05), there was a decline of the orienting-exploratory behavior indicators to control values. This indicates that afobazol administered during the prenatal period and / or entering through the milk postnatally is able to reduce such negative effects of the peat smoke as heightened emotionality and slowing fading orientation and exploratory behavior in the offspring.
