Polyoxometalates antiviral activity and their impact on cytokine-s genes expression

Abstract:
Keggin and Andersen polyoxometalates were investigated in this work. Their cytotoxiс effect on cells was examined by MTT assay. Heteropoly acids with Andersen anions were with greater toxic action on cells than Keggin-s heteropoly acids. Keggin POM-s influence on hemagglutinating and infectious activity of different influenza strains (influenza А/Aichi/1/68(Н3N2), А/California/07/09/(Н1N1) и А/Pert16/09(Н3N2)) was investigated. The most virucidal effect on investigated strains was observed after exposure of Keggin POMs with H4SiW12O40 and H4SiMo12O40. POMs effect on cytokines synthesis was investigated. A wide spectrum of cytokine mRNA activities after POM exposure was examined in cells culture HEF, A549, L41. It was demonstrated that, Keggin-s heteropoly acids activated gene transcription of INF-γ, IL-18, IL-1β, IL-8, TNF-α, which played an important role in antiviral immunity. Nanoparticles H3PМо12040 and H4SiW12O40 activated IL-10 expression in oncologic cell lines А549 and L41. Il-10 regulates humoral immunity and immune humoral response. As a result, cytokines in vivo promote macrophage and cellular components of immune system and play an important role in infectious diseases. Drug development with POM is promising for whether mono- or complex therapy of viral, autoimmune and oncologi-cal diseases.
