Spectrophotometric analysis of phenolic compounds in alcohol infusion from fresh leaves of Hypericum perforatum L

Abstract:
Ethanol infusion of fresh leaves of the plant Hypericum perforatum, prepared by the method used for determination of chlorophyll proved the possibility of determining the content of flavonoids and phenol carbonic acids, as well as hypericins. The content of flavonoids and hypericin can be studied in extracts of fresh leaves of H. perforatum by spectrophotometric methods. The infusion of the fresh leaves contains 20 % more flavonoids than the extract of the dry leaves. Differences in the content of hypericins were insignificant (< 5 % of the values obtained). The definition of the content of phenol carbonic acids in samples of fresh leaves of H. perforatum by direct spectrophotometry was incorrect. The proposed approach can be applied when studying the dynamics of accumulation of phenolic compounds and variability of these compounds in individual plants or small groups (populations) of H. perforatum. The extract obtained can be analyzed by HPLC. Metrological characteristics of the proposed methods of analysis were determined and shown that the accuracy of the determination of the compounds was satisfactory.
