Abstract:
There was carried out the study of the toxic properties of hydrochloride of beta-phenylglutamic acid (neuroglutam) in the dosage form at single intragastric introduction on outbreeded eugamic male and female mice (21-25g) and rats (180-220g). The dosage form of the hydrochloride of the beta-phenylglutamic acid was tested in the form of granulated material, making up the nucleus of the neuroglutame pills and including, as a reactant, the hydrochloride of beta-phenylglutamic acid - 58,82%, as well as polyplasdone XL-10 - 23,53 %, twice-substituted calcium phosphate - 11,01 % and calcium stearate - 0,76 %. There were formed 3 control groups for each species and sex of animals: the 1st - intact, the 2nd group which was getting distilled water; the 3rd group which was getting placebo (the adjuvants of the neuroglutame pills). There was single oral introduction of the granulated material of the neuroglutame pills into the trial groups of animals: to mice in the doses of 5000 mg/kg to 9000 mg/kg; to rats - of 10000 mg/kg to 19000 mg/kg. The calculation of LD50 was conducted by the method of Litchfield and Wilcoxon. The statistic analysis of the dynamics of the body weight of the animals was carried out using Microsoft Excel 2007, Student-s t-criterion. It was ascertained that the LD50 of the granulated material, making up the nucleus of the neuroglutame pills at intragastric introduction constitutes: 6469,06 (5795,76-7220,58) mg/kg in male mice, 7660,45 (7074,46-8294,98) mg/kg in female mice; in male rats - 16154,77 (14387,43-18139,20) mg/kg, in female rats - 13982,32 (12179,20-16052,40) mg/kg, which, according to the klassification of I.V. Sanotskiy allows to associate the stated dosage form to the class of low-toxic agents. In the clinical picture of the intoxication of mice and rats of both sexes prevail the symptoms of deprivation of CNS: reduction of mobility, sedation, ataxia, hyporeflexia, depression of pain sensitivity, periodical generalized tremor, generalized convulsions leading to coma and respiratory depression.
