Selection of solvent to study the impact of water-insoluble substances on the coagulatory component of hemostasis under conditions in vitro during non-clinical stage

Abstract:
The purpose is to study potential solvents of water insoluble substances on the coagulative component of hemostasis under conditions in vitro. Targets of research are ethanol, DMSO (dimethylsulfoxide), DMF (N,N-dimethylfomamide) and dioxan (diethylenedioxide) in analog exposures. Materials and techniques. Experiments have been performed on the blood of healthy male donors. Patterns of non-thrombocytic plasm have been received by spinning citrated blood at 300g during 15 minutes. Coagulative activity of solvents has been defined in vitro by doing standard clotting tests on turbodimetric hemocoagulometer Solar CGL (made in Belarus). Activated partial thromboplastin time, prothrombin time and fibrinogen exposure have been registered according to A.Clauss. The findings have been processed with statistics package Statistica 10,0 (StatSoft Inc, Usa). Normalcy of distribution of actual data has been checked with the help of Shapiro-Wilk statistics. Dispersion analysis has been performed with Kruskal-Wallis criterion. Critical level of significance p for statistic criteria is assumed equal to 0,05. Results. It has been defined that virtually all the researched solvents exert influence on the coagulative component of hemostasis. Our findings define that even 1 % (vol/vol) DMSO water solution shows expressed anticoagulative activity. DMF and dioxan exert almost similar influence on the coagulative component of hemostasis and prolong the running time of all defined clotting tests even in 0,1 % (vol/vol) exposure. Ethanol exerts less influence on coagulation of non-thrombocytic plasm. The findings have revealed such exposures when ethanol does not show anticoagulative activity. The findings show, that among the researched solvents for water insoluble compounds, ethanol is the most physiology-based solvent exerting minimal influence on hemostasis with exposure not exceeding 50 % (vol/vol).
