Analysis of lipophilic extract of the herb Viola tricolor L. by method GC-MS

Abstract:
The lipophilic extract of the herb Viola tricolor L. was analyzed on an mass spectrometry that was performed on an Agilent gas chromatograph-mass spectrometer (GCMS) Model 6890/5973, equipped with a DB-5ms fused silica capillary column having (5 %-Phenyl) ? methylpolysiloxane stationary phase (30 m length ×0.25 mm internal diameter and 0.25 μm film thickness), programmed from 50°C (5 min) to 320 °C at 4 °C/min and held for 5 min. The mass spectrometer (MS) was in electron impact mode at 70 eV and electron multiplier was at 2200 V. Ion source and MS quadrupole temperatures were 230 °C and 180 °C, respectively. Mass spectral data were acquired in the scan mode in the m/z range 33-450. The identification of components in the oil was based on retention indices relatives to n-alkanes and computer matching with the WILLEY 275.L library, as well as by comparison of the fragmentation patterns of the mass spectra with those reported in the literature. The proportions of the identified compounds were calculated by internal normalization. Twenty eight constituents were determined in the herb Viola tricolor L. lipophilic extract. The main constituents were found to be linoleic acid (443.86 mg/kg), palmitic acid (303.26 mg/kg), nonacosane (75.69 mg/kg), 2,3-dihydro-3,5-dioxy-6-methyl-4Н-pyran-4-оn (32.59 mg/kg), γ-sitosterol (23.29 mg/kg), phytol (15.19 mg/kg) and heptacosane (11.98 mg/kg). Among the main constituents of lipophilic extract of the herb Viola tricolor L. have been identified markers for standardization of Viola tricolor L. in medicinal mixtures. As markers of Viola tricolor L. can be used 2,3-dihydro-3,5-dioxy-6-methyl-4Н-pyran-4-оn, γ-sitosterol and phytol.
