Abstract:
In All-Russian Institute of Medicinal and Aromatic Herbs are work out technology of complex preparation - effervescent granules with Malva sylvestris dry extract and Glycyrrhiza glabra L. dry extract. Granules consist from dry extracts, citric acid, sodium hydrocarbonate, microcrystalline cellulose (MCC 90), polyvinylpyrrolidone. Active component of Glycyrrhiza glabra is glycyrrhizic acid, that may be determined by HPLC. The HPLC method with spectrophotometric detector don-t used for Malva sylvestris polysaccharides analysis. The standartization of granules need taked at conention of glycyrrhizic acid. The aim of this study is the choice of conditions for HPLC analysis of glycyrrhizic acid in effervescent granules. The chromatographic system WATERS with spectrophotometric detection at (λ=254 nm), using a C18-column Kromasil C18, 5μ, 250×4.0 mm (Dr Maisch), with flow rate of 1.0 ml/min was used for HPLC analysis. The Glycyrham was used as standart. Glycyrham is monoammonia salt of glycyrrhizic acid. The mobile phases acetonitrile - water (40:60) with 1 % CH3COOH; acetonitrile - water (40:60) with 2 % CH3COOH; acetonitrile - water (40:60) with 5 % CH3COOH; acetonitrile - water (40:60) with 0.5 % H3PO4; acetonitrile - water (40:60) with 1 % H3PO4 were investigated. The mobile phase acetonitrile - water (40:60) with 1 % H3PO4 was choisen as optimal. Were detected, that dependence area of standart peak from concentration are lineac in the range of 0.05-1.0 mg/ml. The standart (glycyrham) and effervescent granules are soluted in solution EtOH - H2O (1:1) before HPLC-analysis. Were determinated that time retention of glycyrrhizic acid is ~ 6.3 min. The time retention of glycyrrhizic acid at chromatograme of standart and at chromatograme of effervescent granules are conflicting. The peak at ~ 6.3 min - is β-epimer of glycyrrhizic acid. The peak at ~ 9.2 min - is α-epimer of glycyrrhizic acid.
