Xanthine oxidase as enzymatic biotest-system for in vitro revealing of biologically active substances possessing anti-inflammatory and anti-arthritis properties

Abstract:
We used an enzyme xanthine oxidase (XO) to generate an enzymatic biotest-system for in vitro screening of biologically active substances (BAS) with anti-inflammatory and anti-arthritis activity. Enzymatic activity of XO was determined using of formed uric acid at catalytic oxidation of xanthine Effect of BAS was estimated to reduce the enzymatic activity of the XO. Proofs of legitimacy of application xanthine oxidase as test object for BAS identification with anti-inflammatory properties are provided. Verification of XO-biotest -system made with a specific XO inhibitor (allopurinol) and known drugs with proven anti-inflammatory and anti-arthritis activity (diclofenac, voltaren, meloxicam, ketoprofen, nimesulide). Proofs of legitimacy of application xanthine oxidase as test object for BAS identification with anti-inflammatory properties are provided. Testing conducted with KO-test system for BAS related to other pharmacological groups (antimicrobic, antiviral and gepatoprotector drugs, neuroprotectors, antioxidants, adaptogens, thyreo- and cardiotropic means) The presence of anti-inflammatory properties in a number of antimicrobial and antiviral agent were found out (sangviritrin, alpisarin, hyporamin, helepin D, saliphozid). It was shown that neuroprotectors pyracetam and patrinosid, antioxidants mexydol and mexamin, hepatoprotector silibin, thyreo- and cardiotropic BAS extracts zuznik and laptchatka were possessed of anti-inflammatory properties. It was shown that adaptogen zhenshen practically didn't influence on XO activity. The obtained data testify that XO-test- system can be applied as a way of preliminary revealing in vitro anti-inflammatory and antiarthritic properties of BAS.
