Abstract:
The gastro-protective activity of the grape leaves red extract dry (GLRED) has been evaluated in white non-linear male rats in experimental models of stomach ulcers induced by 96 % ethanol (1 ml/rat, decapitation after 1 hour) and induced by indometacin (30 mg/kg, decapitation after 24 hour). GLRED was administered in concentration 10 mg/kg and 100 mg/kg. Ranitidine as comparative drug in concentration 100 mg/kg was administered. GLRED and Ranitidine were introduced intraperitoneally for 3 days. Stomach and duodenum of rats after decapitation were extracted and quantity of ulcers and ulcer area (by microscope MBS-10), Index Paulus (IP) and Therapeutic Effect (TE - ratio of experimental IP to control IP) were calculated. The Student t-test was used for estimation of statistical confidence.
Significant gastroprptective effect was found after introduction of GLRED (10 mg/kg) ? on experimental model of ulcers caused by ethanol area of gastric ulcer decreased by 78,6 % (TE=4,7) and on indometacin-induced ulcer area of gastric ulcer decreased by 45,0 % (TE=1,8, p < 0,05). Introduction of GLRED (100 mg/kg) reduced the gastric ulcer area by 92,2 % (TE=12,9, p < 0,05) ? ehanol-induced ulcers, and by 81,2 % (TE=5,3) ? indometacin-induced ulcers. Thus, marked dose-dependent gastroprotective effect of GLRED was found. The reliable gastro-protective effect of Ranitidine on indometacin-induced model of stomach ulcers was found - ulceration reduced by 58,4 %, TE = 2,4. Decrease of ulceration after introduction of Ranitidine on ethanol-induced model of stomach ulcer was insignificant (decrease by 10,5 %, p > 0,05). It was found that gastro-protective activity of GLRED (100 mg/kg) exceeded the activity of Ranitidine (100 mg/kg) 2,2 times on indometacin-induced ulcers and 11,7 times ethanol-induced ulcers.
