Development of methods to standardization of Quercus Robur L. in multicomponent plant composition for maintenance ellagic acid using HPLC method

Abstract:
Determined marker for qualitative and quantitative standardization of Quercus robur L. bark in the plant mixtures. As a marker of Quercus robur L. bark can be used ellagic acid. Ellagic acid has a wide range of biological activity (in particular, ellagic acid has anti-inflammatory, hepatoprotective, antioxidant, anti-cancer and many other pharmacological properties). The method for the analysis of ellagic acid in bark and herbal mixtures Quercus robur L. bark using HPLC was developed. Chromatographic study investigated extracts and reference solution of ellagic acid was carried out by chromatograph Shimadzu ser. 20, equipped with a diode array detector under the following conditions: column Phenomenex Luna C18 (2) (250 mm × 4.6 mm, 5 micron particle size); the column temperature - 35 ºC, detection wavelength - 330 nm, the flow rate of the mobile phase - 1ml/min volume injection - 5 mcl. As the mobile phase used a 0.1 % solutions of trifluoroacetic acid in a mixture of water and acetonitrile (gradient elution). With the developed methodology analyzed 5 series Quercus robur L. bark. The analysis shows, that the content of ellagic acid in raw plant is between 0,0439 ± 0,0023 % to 0,0787 ± 0,0032 % in terms of the dried materials. It is shown, that the presence and content of ellagic acid Quercus robur L. bark can be standardized in the mixtures with the following plant: leaves of Urtica dioica L., roots of Cichorium intybus L., seeds of Linum usitatissimum L., roots of Curcuma longa L., roots of Acorus calamus L., roots of Zingiber officinale Rosc. and seeds of Avena sativa L.
