Development of qualitative detection and quantitation methods of the sum total of flavonoids in the collection «Lorpoliphyt»

Abstract:
For treatment of inflammatory diseases of the upper air passages (UAP) there has been offered the seven compound officinal vegetative collection: plantain leaves, chamomile flowers, marigold flowers, equisetum herbs, nosebleed herbs, hypericum herbs, elecampane roots and rhizomes. The flavonoids containing in the marigold, the equisetum, the camomile, the hypericum and the nosebleed cause the anti-inflammatory effect which is the basic one in the herbal medicine of UAP diseases. The aim of research is working out of techniques of qualitative detection and quantitation of the sum of flavonoids in an officinal vegetative collection. The thin-layer chromatography method is put in a basis of a technique of qualitative detection of the sum of flavonoids. There has been developed the technique TLC on plates «Sorbfil PTSH-AF-A-UF» in the system of solvents n-butanol - acetic acid - water (4:1:5), zones of adsorption of aluminum have been revealed by chloride. The sum presence of flavonoids in investigated collection has been established. The method of quantitation of total flavonoids in the collection is based on the method of differential spectrophotometry. As a standard substance they ahve selected rutin. The optimal conditions for flavonoids extraction from collection are: chopped raw material of 1 mm, the extractant 70 % alcohol, the ratio of raw material and extractant 1:50, extraction time 45 min and 15 min in a boiling water bath under reflux. Based on the given research, they have developed a method which consists in carrying out of the reaction of complexation of flavonoids with aluminum chloride and measuring of the optical density of extraction from the sample collection relative to the reference solution (DLS rutin) with a spectrophotometer at a wavelength of 407 nm. The reliability of the spectrophotometric determination results of total flavonoid content in the test collection in terms of the rutin has been tested by supplementation method. The relative error of 0.95 at a confidence level does not exceed ± 2.57%. The content of sum total flavonoids in the collection ranges from 1.36 to 1.65%.
