The IGF-System as One of the Pathogenetic Factors and Potential Target for Antitumor Therapy or Prostate Cancer

Abstract:
The authors discuss recent 10-years data about mechanisms of realization of IGF-signaling pathway effects on prostate cells. The IGF-signaling pathway is a multicomponent system including IGF, IGF receptors and IGF binding proteins, and acting on cells in endocrine, and also auto/paracrine manner. The data of metaanalysis of clinical significance investigations demonstrate undoubted involving of IGF, IGF-R1 and IGFSB in prostate cancerogenesis and tumour progression, although precise correlations between components of IGF-system and prostate cancer rescue, clinico-morphological features and prognosis in prostate cancer are not established because of complete cross-interactions with other mithogenic and proapoptotic signaling pathways. Different components of IGF-system could be interesting as potential targets of antitumour therapy of prostate cancer: there are more perspective data about antitumour effects of antiIGF-R1 monoclonal antibodies, and of tirosinkinase inhibitors.
