Abstract:
Pharmacokinetic parameters of phenibute substance at intravenous injection and oral intake in extra therapeutic dose 50 mg/kg have been studied. Phenibute was tried to be found in blood, heart, brain, lungs, spleen, muscles, omentum and in organs, providing elimination - a liver and kidneys, as well as in urine and excrement. Blood and organ sampling at intravenous injection was produced in 5, 10, 20, 40 minutes, 1, 2, 4, 8, 12 h; at oral intake - 15, 30 minutes, 1, 2, 4, 8, 12 h after the drug injection and in 5 minutes in animals test group. It is shown up, that the maximum amount of phenibute were found in the lungs, spleen, liver and kidneys, and the minimum one - to muscular tissue and brain. Drug pharmacokinetic profile in heart coincides with such in blood plasma. Phenibute distribution in organs and tissues at oral intake in general is similar to such at intravenous injection. Drug absolute bioavailability is 43,6 %, maximum excretion is mainly performed via kidneys during the first 24 hours after intake.
