Comparison of neurotropic effect di- and tetramethylderivatives dihydroisoquinolilaminocaproic acids

Abstract:
Neurotropic effect of di- and tetramethylderivatives of 3,4-dihydroisoquinolin-1-ylaminohexanoic acid has been studied in mice by hot plate, open field, elevated plus-maze and light/dark box tests. The presence of additional methyl groups in position 4 of isoquinoline had changed compounds - pharmacological profile significantly. 6-(3,3-dimethyl-3,4-dihydroisoquinolin-1-yl)aminohexanoic acid (compound I) have had central analgesic effect in dose-dependent manner, its ED50=13,5(9,5-19,2) mg/kg in the hot plate test. Сompound I has exhibited no depressant influence on central nervous system. 6-(3,3,4,4-tetramethyl-3,4-dihydroisoquinolin-1-yl)aminohexanoic acid (compound II) has been ineffective in analgesic activity but produced anti-anxiety effect with stimulant component. Both substances are considered as promising neurotropic agents.
